waiting...
I made movies of 11 strains of PTEN- mutants this morning, and I have 13 other movies to make of my HO46 or WT cells. One of the cell lines is Alex’s HO46, because his appear to move slower than mine, ryans, ethans, and allens. We want to see if the problem is with the cells or his handling.
I need to leave by 5:30 today, so I decided to make third movies of my cells that look obviously...

So yesterday, while looking at my fruiting bodies, I found out that almost all of my plates from one transformation exhibited a phenotype similar to P13k1/2- mutants. I remember plating my HO12 PTEN- mutants on Ka labeled P13K1/2 because my flask of Ka labeled PTEN disappeared…and that shouldn’t have done anything unless that flask of Ka was contaminated with P13K1/2- mutants. The past week has been crazy! Assays assays assays. The only times I’m ever in the cave anymore are ten o’clock meetings with Delquin, and when I’m running my movies and analyzing my data on MATLAB.
All other time is spent in the Weiner Lab splitting cells, freezing down cells, plating tens of fruiting bodies, preparing cells for movie assays, the occasional transformations when...

So while freezing down cells today, I spilled an entire 50ml tube of freezing media on myself. I felt horrible. That thing must have costed much $$$. And as if it couldn’t get any worse, I felt stinging on my legs. I looked down, and realized that the media had soaked through my jeans in a few choice areas. Great. SO TIRED! From waking up at ~6:50 everyday to catch the 8:00 shuttle to leaving at 7:50, 8:30, 6:00…UCSF being my second home isn’t a joke anymore, it’s real! But it’s all good, I like it here :)

Day...6?
I was suppose to finish blogging friday, but my bed looked so inviting I thought I would lie down for a moment. Bad mistake. I woke up at ten? eleven? and promptly went back to sleep after a light dinner. I guess by the end of bootcamp and my first week at iGEM I was more exhausted than I thought. But it’s a good exhaustion, like after running a lot.
And before I knew it, Monday smacked me...

Day Three
I started my day off with a chocolate cookie with the hopes of staying awake and alert during today’s lectures; I failed. I don’t know why, but it was extraodinarily difficult today. It wasn’t the topics or the speakers, but me…I think I need more sleep the night before. And I need to learn how to manage my time better - aka start my hw earlier. I think I should do hw now...
